Lipidomics
Metabolomic ServiceLipidomics analyses cover fatty acid-derived hydrophobic metabolites that are not or only incompletely included in metabolomics panels. By extraction with unpolar solvents, a wide range of lipid species (Tri-/diacylglyceroles, glycerophospholipids, sphingolipids, cholesterol esters etc.) become amenable to mass spectrometry-based targeted lipidomics methods. Visit the section "Lipidyzer", for more information about our Lipidomics assay.
We furthermore offer targeted lipidomics analyses of pro-inflammatory and pro-resolving lipid mediators (Prostaglandins, Leukotrienes, Lipoxins, Resolvins etc.) that are relevant in inflammatory processes. More information can be found under the section "Eicosanoids".
We use the ilab Core Management system to book services by internal and external users.

Our platforms
The Lipidyzer/Shotgun Lipidomics is a targeted Lipidomics platform that allows accurate quantification of up to 1070 lipid species from 13 lipid classes using ion mobility mass spectrometry (IM-MS) with multiple reaction monitoring (MRM). Please note, that the actual number of lipid species that can be quantified is dependent on sample matrix and sample quality. In general, between 500 and 800 lipids are reliably quantified. The lipid quantification is based on known concentrations of internal standards (approx. 10 different fatty acids with various amount of carbon atoms and double-bonds per lipid class) that are spiked into the analytical samples prior the extraction procedure. The concentration values are reported as nmol/mL for liquids and nmol/g for tissues, respectively.
- Types of samples measured: plasma, serum, cell culture, brain, heart, kidney, lung, muscle, liver, white adipose tissue, brown adipose tissue
- Amount of sample requested for plasma/serum: 35 µl (better 50 µL); tissue: between 20-30 mg dependent on matrix
- Species tested: human and mouse

The UHPLC-MS/MS method contains 58 oxylipins and PUFA precursors covering pro-inflammatory and pro-resolving lipid mediators from all known major pathways, i.e. COX- , LOX-, and CYP450 mediated lipid conversion, as well non-enzymatic oxidation. The identification and quantification is achieved by retention time and multiple reaction monitoring (MRM), including 9-point calibration with 17 isotopically labelled internal standards.
