mRNA Reprogramming
Target of this service is the generation of the iPSCs from adherent somatic cells (e.g. dermal fibroblasts) using a non-integrative mRNA-based method.
Please email the completed submission form and provide a health certificate outlining that the health status of the donor or cryopreserved cells is pathogen free (HIV-, HBV-, HCV-). The primary somatic cells must be mycoplasma negative.
Sample submission: Fresh/cryopreserved skin biopsy or cryopreserved adult/neonatal primary somatic cells:
- The primary somatic cells should be at a low passage (passage 8 or lower).
- The culture should exhibit a rapid proliferation rate and not show signs of senescence.
- Primary cells will be processed for reprogramming unless the cultures exhibit signs of senescence or abnormal morphology.
The service requires approximately 2-3 months for completion.
Cells are plated on Matrigel matrix and transfected daily with mRNA cocktail.
Three clones exhibiting the best morphologies and growth rates are further expanded for 4-5 passages and cryopreserved (2-5 vials per line).
Clones are shipped to the client as frozen vials.
Remark: The reprogramming method is also suitable for immortalized fibroblasts.
