mRNA Reprogramming

Target of this service is the generation of the iPSCs from adherent somatic cells (e.g. dermal fibroblasts) using a non-integrative mRNA-based method.

Please email the completed submission form and provide a health certificate outlining that the health status of the donor or cryopreserved cells is pathogen free (HIV-, HBV-, HCV-). The primary somatic cells must be mycoplasma negative.

Sample submission: Fresh/cryopreserved skin biopsy or cryopreserved adult/neonatal primary somatic cells:

  1. The primary somatic cells should be at a low passage (passage 8 or lower).
  2. The culture should exhibit a rapid proliferation rate and not show signs of senescence. 
  3. Primary cells will be processed for reprogramming unless the cultures exhibit signs of senescence or abnormal morphology.

The service requires approximately 2-3 months for completion.

Cells are plated on Matrigel matrix and transfected daily with mRNA cocktail.

Three clones exhibiting the best morphologies and growth rates are further expanded for 4-5 passages and cryopreserved (2-5 vials per line). 

Clones are shipped to the client as frozen vials.

Remark: The reprogramming method is also suitable for immortalized fibroblasts.
