Sendai Reprogramming
Aim of this service is the generation of iPSCs from PBMCs isolated from whole blood using Sendai virus.
Sample submission: fresh whole blood/ freshly isolated PBMCs from whole blood. The blood sample must be pathogen free (HIV-, HBV-, HCV-).
The cryopreserved PBMCs will be processed for reprogramming unless the culture exhibits high percentage of cell death.
The service requires 4-6 months for completion.
Cell are transduced with Sendai virus and the reprogramming will be performed under feeder-free conditions.
Single-cell clones exhibiting the best morphologies and growth rates are further expanded for testing Sendai virus absence (TaqMan-based RT – PCR assay) and for cryopreservation.
Two Sendai negative clones are shipped to clients as frozen vials (5 vials per clone).
Sendai negative clones will be declared as BSL-1 (S1). The iPSCs are genetically modified organisms by definition despite the lack of foreign DNA, therefore customers require BSL-1/S1 laboratory for storing and handling the cells according to GenTSV.
